Chinese and American researchers reported in a new issue of the journal Science Translational Medicine on the 14th that a new imaging technique called stimulated Raman scattering microscopy can quickly and accurately detect brain tumor tissue, thus helping surgeons to be safer. The resection surgery is performed effectively.

The first author of the paper, Professor Ji Minbiao, who is currently working in the Physics Department of Fudan University, told Xinhua that the brain surgeons can only rely on the experience of the naked eye and touch to determine the tumor boundary during surgery, and their technology can help doctors during surgery. Quickly identify residual tumors, improve surgical accuracy, and achieve precise treatment.

The study was the result of a collaboration between Dr. Daniel Olinger of the University of Michigan Medical School during his postdoctoral fellowship at Harvard University. Ji Minbiao said that stimulated Raman scattering microscopy mainly uses tissue cell density, axon density and lipid protein ratio to identify the degree of tumor infiltration. Tests on the tissue of 19 brain tumor patients showed that the sensitivity and specificity were 98%.

Compared with other brain tumor detection techniques, the new technology has two major advantages: First, it is a label-free technology that does not require the introduction of markers such as dyes, fluorescent molecules or fluorescent proteins, and can directly detect the spectral signal of the sample itself; It can distinguish and detect them according to the spectral differences between various biomolecules, such as lipids, proteins and nucleic acids, and has good molecular specificity.

According to Ji Minbiao, the first in vitro tissue imaging system based on stimulated Raman technology has been applied in the brain surgery operating room of the University of Michigan Medical School, so the detection of isolated surgical samples is “very close”. However, it is necessary to use a certain distance in the body detection, mainly the consideration of the laser safety dose, as well as the design and manufacture of the hand-held probe, and the limitation of the size of the imaging field to be solved.

Stimulated Raman technology is also emerging in China. At present, the Biodynamic Optical Imaging Center of Peking University and Fudan University are engaged in related research and development.
